It is necessary to use an efficient delivery system to enhance drug delivery to intended cells and reduce side effects of cytotoxic agents. In this study, a cancer-specific aptamer was adopted for cancer-directed delivery of nanoparticles containing anti-cancer siRNAs. Aptamers made of nucleic acids have been developed to have targeting capability to certain specific molecules with strong physical stability. In addition, they are smaller and less immunogenic compared to antibody molecules. Aptamer-conjugated liposomes exhibited a tumor-specific binding and delivery capability. To prove a potential application of aptamer-conjugated liposomes in cancer therapy, anti-EGFR aptamers were conjugated to liposomal nanoparticles encapsulating anti-tumoral siRNA and then compared to anti-EGFR antibody (cetuximab)-conjugated liposomes in terms of tumor-specific binding and siRNA delivery. A431 cells (EGFR-positive) and HepG2 cells (EGFR-negative) were transfected with the two different systems and then analyzed by confocal microscopy and FACS.
